Abstract : Two Red Sea’s soft coral animals identified as Rhytisma fulvum fulvum and Xenia umbellate were collected off Jeddah coast. Both organisms were separately extracted with organic solvents and fractionated and purified using different chromatographic techniques including column chromatography and thin layer chromatography. The purified secondary compounds were identified with the aid of 1D and 2D NMR spectroscopy involving (1H, 13C, COSY, DEPT, HSQC, NOESY, and HMBC) in addition to IR spectrum and MS spectrometry. From the soft coral R. fulvum fulvum four compounds, including 12-O-acetyl-nardosinan-6-en-1-one (40) and 6β-acetyl-1(10)-a-13-nornardosin-7-one (41) are described as derivatives nardosinane type sesquiterpenes, and known sesquiterpenes 4-acetoxy-2,8-neolemnadien-5-one (45), and nephthenol (46) a known cembranoid diterpene. Compounds 40, 41, 45, and 46 showed cytotoxic effects against tested cancer cell lines (prostate cancer, PC-3; human liver carcinoma, HepG2 and human colon adenocarcinoma HT-29). Compound 40 showed highest activity against HepG2, HT-29, and PC-3 with IC50 values of 9.7±1.13, >100, and 8.9±1.15µg/ml respectively. Compound 41 showed against HepG2, HT-29, and PC-3 with IC50 values of 38±4.1 >100, and 5.5±0.61µg/ml, respectively. Compound 45 showed IC50 values of 8.5±0.91, >100, and 7.4±0.85 µg/ml, respectively, also compound 46 showed IC50 values of >100, 9.1±1.01, and >100 µg/ml respectively. Xenia umbellate yielded six compounds 3β-,5α-,6β-,11α-,20β-pentahydroxygorgosterol (52) and 23,24-dimethylcholest-16-ene-3β,5α,6β,11a,20(R)-pentol 3-monoacetate (97) known as steroids, xeniumbellal (53) known is a diterpenoid, aromadendrene (54), palustrol (55) and viridiflorol (56) known as sesquiterpenes. The cytotoxicity potential of the isolated compounds 52, 53, 54, 55, 56, and 97 were tested against HepG2, HT-29, and PC-3 cancer cells. Compound 52 showed cytotoxic activity with IC50 values of 2.75±0.12, 9.30±0.9 and >100 µg/ml, respectively. Compound 53 showed cytotoxicity with IC50 values of 5.7±0.5, 3.8±01, and 1.7±0.1 µg/ml respectively. Compound 54 exhibited activity with IC50 values of 2.3±0.14, 1.69±0.19, and 3.1±0.27 µg/ml respectively. 55 showed cytotoxicity against HepG2 with an IC50 value of 2.5±0.09 µg/ml and against PC-3 with an IC50 value of 15.3±0.171µg/ml. 56 exhibited cytotoxicity effect with IC50 values of 3.09±0.09, >200, and 2.90±0.59 µg/ml, respectively. 97 showed cytotoxic effect with IC50 values of 4.80±0.019, >100, and 5.65±0.59 µg/ml, respectively.
